Location on barley chromosome 1 of loci Estl and Est2, coding esterases 1 and 2

The leaf esterases were analysed by using polyacrylamide gel electrophoresis, pH 8.3 (Netsvetaev, 1992a,b). Esterases 1, 2, 4, and 5 were identified by means tester set of varieties (Table 1) received from H. B. Johansen in 1989. The data of addition wheat-barley lines (obtained from A. K. M. Islam in 1988) analyses confirmed that Estl, Est2, and Est4 are located on barley chromosome 3 and EstS on chromosome 1.

Differences in esterase isozymes composition between varieties having the fastest and slowest electrophoretic mobility were controlled by new loci designated as Estl l and Estl2, respectively.

Recombination value frequencies between genes Estl, Est2, f2 (chlorina 2), al (albino lemma), cu2 (curly 2), and uz (semi-brachytic) were calculated on results of the cross cu × al+f2. P1 (=cu) had alleles F2, Al, cu2, uz, Est1Af, est2 (absent esterase 2 activity, and P2 (=al+f2) had alleles f2, al, Cu2, Uz, Est1Ca, Est2. The data obtained is presented in Table 2.

The arrangement of these six genes on chromosome 3 are as follows

and are shown in Figure 1.

Loci Estl - Est2 are tightly linked and no recombinants in this region were found.

The information reported here is additional to data previously obtained by Nielsen and Hejgaard (1985), Konishi and Matsuura (1989), and Netsvetaev (1992b) concerning Est location on chromosome 3.
