II. 11. Telotrisomic analysis in Triplo 3S in barley. (1)

A. Shahla and T. Tsuchiya, Department of Agronomy, Colorado State University, Fort Collins, Colorado 80523 U.S.A.

(1) Supported partly by USDA/SEA Competitive Grant 5901-0410-9-0334-0 and 82-CRCR-1-1020 to T. Tsuchiya.

Two marker genes als (absent lower laterals) (Kasha and Walker, 1960) and gs2 (Glossy sheath 2) (Robertson et al. 1965) were used in telotrisomic analysis with Triplo 3S. This experiment was conducted to find the definite arm location of both marker genes on chromosome 3 of barley. Crosses were made between telotrisomic 3S plants and double marker stock als gs2. The segregation ratios were observed in the F2 generation (Table 1).

Table 1. F2 data for telotrisomic analysis of double marker stock als gs2 with Triplo 3S.

The segregation ratio in F2 generation fits well to a 3:1 disomic ratio. Also the presence of appreciable number of homozygous recessives in trisomic portion indicates that both genes are located on the opposite arm (3L). These results are in agreement with the previous arm location of both genes in linkage maps of barley (Tsuchiya and Singh, 1982).
