Abstract
Wheat ( Triticum durum ) samples from the USA, submitted for Tilletia indica quarantine surveys, were also analysed using the blotter test method to monitor the kernel mycoflora. The fungi were identified based on colony characteristics on the kernels and/or on artificial media. Molecular techniques using internal transcribed spacer (ITS) region universal primers and GeneBank research by BLAST were used to find similar sequences. The results indicated the presence of Fusarium verticillioides , Fusarium proliferatum , Bipolaris sorokiniana [ Cochliobolus sativus ], Bipolaris spicifera [ Cochliobolus spicifer ], Drechslera japonica [ Pyrenophora japonica ] and Exserohilum rostratum . With the exception of Bipolaris heveae and B. ellisii [ Cochliobolus ellisii ], the other fungal species observed were already present in Italy. The usefulness of seed and crop monitoring to avoid the introduction of new pathogens into the country, even if not under quarantine regulations, was also discussed
