Abstract
The immature embryo of beer barley cultivar Jinyin 6 was used as biolistics transferring acceptor. The plasmids with thioredoxin a gene (target gene) and bar gene (selective gene) were co-transformed into barley through particle bombardment. Selection, regeneration and rooting were conducted on media containing 3-5 mg/litre PPT. The effective method of transformation and plant regeneration was identified. Results indicated that genetic transformation efficiency can be improved using immature embryos pre-cultured for 15 days. Culturing callus on media containing ABA [abscisic acid] at 1 mg/litre for 2 weeks resulted in the inhibition of immature embryo germination and improvement in embryonic callus development. The efficient plant growth regulator proportions for bud differentiation were zeatin at 1 mg/litre + IAA at 0.1 mg/litre. Eleven regenerated plantlets were obtained from 178 PPT resistant calluses with a regeneration frequency of 6.2%. The Trxs gene in the plants under different treatments were assayed into barley genomes
